Twist Bioscience Faces a Lofty Bar After a 72% Rally on AI Drug-Discovery Momentum

Twist Bioscience heads into its fiscal fourth-quarter report having raised its full-year revenue outlook three times, promised a breakeven quarter on adjusted EBITDA, and watched its stock surge roughly 72% since the last report while the S&P 500 barely moved. That combination makes Thursday morning's release less about whether the company can hit its numbers and more about whether its AI-enabled drug discovery story can justify a valuation that has already priced in a lot of good news.

The Street is looking for a loss of $0.44 a share on revenue of $123.4 million, which would mark growth of nearly 25% from the year-ago quarter. That top-line forecast sits squarely inside management's guidance of $123 million to $124 million, a range that was itself well above where consensus stood before the company spoke last quarter. On earnings, the expected improvement is marginal, roughly 2% better than last year's loss of $0.45, though the whisper number of a $0.40 loss suggests some investors expect cost discipline to show through. The gap is modest, but it hints that the real bar is a slightly narrower loss rather than simply matching the consensus.

The pattern of guidance matters here. Management lifted its fiscal 2026 range from an initial $425 million to $435 million all the way to $456 million to $457 million, and last quarter's raise was about three times the size of the beat. A company that has trained investors to expect upside will be judged on whether it clears the top of its own range, not the midpoint. A result near $123 million, while technically on plan, could read as a pause in momentum.

The operational heart of the report is therapeutics and AI-enabled discovery. Therapeutics revenue grew 49% last quarter to $40.4 million, and management not only said it expects to meet or exceed triple-digit AI order growth this year but also extended that goal into fiscal 2027. The reasoning was that AI customers have shifted from lumpy, model-building orders to smaller, more frequent repeat orders, which should make demand more visible. Continued growth in the 40% to 50% range in therapeutics, along with commentary reaffirming the fiscal 2027 target, would validate that thesis. Any sign that AI orders are reverting to lumpiness would undercut the central reason the stock has rerated.

Several secondary threads also need resolution. NGS applications slowed to 12% growth last quarter, and management pinned a rebound to above 20% on a diagnostic customer ramp, so that is a testable promise. Academic and government demand snapped back to 32% growth after a near-flat first half, and investors will want evidence it was not a one-quarter catch-up given an uncertain funding backdrop. The DNA synthesis and protein solutions business showed softer sequential dynamics as mix shifted toward shorter antibody sequences, while peers are reportedly discounting. Early traction from the newly launched complex genes offering would help offset those concerns. On profitability, gross margin of 52.8% and 70% incremental drop-through set up the adjusted EBITDA breakeven goal, and a roughly $5 million operating expense reduction should help. Missing breakeven after flagging it so clearly would sting.

Sentiment has improved from barely bullish to moderately so, and the stock at $140.30 sits well above its 200-day average of $89.71. Still, shares are far below the post-earnings high of $219.71, meaning the run included a sharp pullback and plenty of volatility. Ultimately, the report will hinge on whether Twist can show that AI-driven therapeutics demand is becoming a recurring, visible revenue stream while it crosses into EBITDA breakeven. If both hold and fiscal 2027 commentary stays firm, the growth narrative remains intact; if either wobbles, a stock this extended has little cushion.
